New enantiopure P,P-bidentate bis(diamidophosphite) ligands. Application in asymmetric rhodium-catalyzed hydrogenation

Two series of new enantiopure bidentate bis(diamidophosphite) ligands with diazaphospholidine and diazaphosphepine heterocyclic backbones were prepared. The ligands have a highly modular structure, which is well suited to the synthesis of a small library of compounds. Preparation was accomplished by the successive addition of enantiomerically pure substituted diamines (N,N?-dibenzylcyclohexane-1,2-diamine (1), N,N?-dimethylcyclohexane- 1,2-diamine (2), and N,N?-dimethyl-1,1?-binaphthyl-2,2?- diamine (3)) and enantiomerically pure diols (butanediol (a), cyclohexanediol (b), di-O-isopropylidenethreitol (c), and binaphthol (d)) to phosphorus trichloride. The corresponding bis(diamidophosphite) selenides were prepared, and the 1JPSe values were calculated in order to evaluate the sigma-donor ability of the new ligands. The cationic Rh(I) complexes [Rh(COD)(P,P)]BF4 were synthesized with 8 of the 12 new bis(diamidophosphite) ligands. The complexes were used as catalytic precursors for the asymmetric hydrogenation of benchmark substrates, namely methyl alpha-acetamidoacrylate (4), methyl (Z)-alpha-acetamidocinnamate (5), and dimethyl itaconate (6). The influence of the nature of both the terminal and bridging fragments of the bis(diamidophosphite) ligands on the asymmetric induction is discussed. Most proved to be effective catalysts for the process, attaining total conversion and excellent enantioselectivity (>99% ee) with the complex containing the (R;Ral,Ral;R)-3c ligand in the hydrogenation of the three substrates. The best performing catalytic precursor [Rh(COD)((R;Ral,Ral;R)-3c)]BF4 was tested in the hydrogenation of selected cyclic enamides (7-9) and beta-acetamidoacrylate (10).

Eleven new chiral macrocycles (1-11, see Figure 1) of the pyridino-18-crown-6 type have been prepared.Nine diazapyridino-crown ligands contain two amide (1, R = benzyl; 4, R = phenyl), two N-methylamide (7, R = phenyl), two thionoamide (2, R = benzyl; 5, R = phenyl), two N-methylthionoamide (8, R = phenyl), two amine (3, R = benzyl; 6, R = phenyl), or two N-methylamine (9, R = phenyl) groups incorporated into the macroring.The appropriate chiral diamine was treated with dimethyl 2,6-pyridinedicarboxylate (or 2,6-pyridinedicarboxyl dichloride),O,O’-dimethyl 2,6-pyridinedicarbothioate, or 2,6-pyridinedimethyl ditosylate to prepare these materials.The macrocyclic diamides were also converted to the macrocyclic dithionoamides using Lawesson’s reagent and the latter macrocycles were reduced to the diamines.A new symmetrically substituted dimethylazapyridino-18-crown-6 ligand (10) and its N-acetyl derivative 11 were also prepared.The interactions of some of the new chiral ligands with (R)- and (S)-ammonium perchlorate were studied by 1H NMR spectral techniques.The degree of enantiomeric recognition was determined by the difference of the free energy of activation values (DeltaDeltaGexcit.) and the difference in log K values for these interactions.The X-ray analyses of the dithionoamido ligands (2, 5, and 8) showed severe deviations of the S and N atoms from the plane of the pyridine ring, especially in the case of 8.The optical rotations of 8 changed with time due to conformational changes.The relevant conformations of 8 are discussed in light of the X-ray crystallography, molecular mechanics, and 1H NMR spectra.

Herein, a dimeric titanium cluster, Ti2Cl2(OPri)6¡¤2HOPri (Ti2), is used in the Claisen-Schmidt condensation reaction, for the selectively activation of symmetrical ketones containing alpha,alpha?-methylene groups and production of alpha,alpha?-bis-substituted alkanones in high efficiency and chemoselectivity. The high efficiency and chemoselectivity can be extended to a variety of typical alkanones and aromatic aldehydes. Both of the oxo-bridged dimeric motif of Ti2 and the ionic Ti-Cl bond are responsible for the high efficiency and chemoselectivity.
