Physicochemical characterization and quality of Dangshan pear wines fermented with different Saccharomyces cerevisiae

Three commercial yeasts strains, namely, Saccharomyces cerevisiae SY, DV10, and Drop Acid Yeast, were used for Dangshan pear wine fermentation. Monitoring main physical and chemical indexes and scoring comprehensive sensory characteristics to find a suitable yeast to produce Dangshan pear wine. The fermentation cycle of SY was short (15?days), and the SY-fermented wine had a suitable sugar-acid ratio, with a residual sugar content of 3.13?¡À?0.05?g/L, total acid content of 3.40?¡À?0.11?g/L, and ethanol content of 14.1?¡À?0.27% (v/v). Additionally, 42 flavor compounds were detected in fermented Dangshan pear wine, and the total amount of flavor compounds was highest in the SY wine (2,584.72?mug/L). Combined with the comprehensive sensory evaluation scores, these results suggest that Saccharomyces cerevisiae SY was the most suitable strain to produce Dangshan pear wine. Practical applications: In this study, we compared the physical and chemical indicators of pear wine brewed by different Saccharomyces cerevisiae in the process of fermentation and the final quality of pear wine products. It was concluded that the pear wine produced by Saccharomyces cerevisiae SY had good quality. The study found a strain suitable for the fermentation of pear wine and provided a theoretical basis for the industrial production of pear wine. Next, we can try to use Saccharomyces cerevisiae SY for large-scale production of pear wine and try to sell it on the market.

Determining the absolute stereochemistry of secondary/secondary diols by 1H NMR: Basis and applications

The absolute configuration of 1,2-, 1,3-, 1,4-, and 1,5-diols formed by two secondary (chiral) hydroxy groups can be deduced by comparison of the NMR spectra of the corresponding bis-(R)- and bis-(S)-MPA esters. The correlation between the NMR spectra of the bis-ester derivatives and the absolute stereochemistry of the diol involves the comparison of the chemical shifts of the signals for substituents R1/R2 and for the hydrogens attached to the two chiral centers [Halpha(R1) and Halpha(R2)] in the bis-(R)- and the bis-(S)-ester and is expressed as Deltadelta.RS Theoretical calculations [energy minimization by semiempirical (AM1), ab initio (HF), DFT (B3LYP), and Onsager methods, and aromatic shielding effect calculations] and experimental data (NMR and CD spectroscopy) indicate that in these bis-MPA esters, the experimental DeltadeltaRS values are the result of the contribution of the shielding/deshielding effects produced by the two MPA units that combine according to the actual stereochemistry of the diol. The reliability of these correlations is demonstrated with a wide range of diols of known absolute configuration derivatized with MPA and 9-AMA as auxiliary reagents. A simple graphical model that allows the simultaneous assignment of the two asymmetric carbons of a 1,n-diol by comparison of the NMR spectra (Deltadelta RS signs) of its bis-(R)- and bis-(S)-AMAA ester derivatives is presented.

Fusaricates H-K and fusolanones A-B from a mangrove endophytic fungus Fusarium solani HDN15-410

Seven compounds including four undescribed fusaric acid derivatives, namely fusaricates H-K, and two undescribed gamma-pyrone derivatives, named fusolanones A-B, as well as a known compound fusaric acid, were isolated from a mangrove endophytic fungus Fusarium solani. Fusaricates H-K represent the first cases of fusaric acid butanediol esters and are diastereoisomers. Synthesis and 13C NMR spectroscopy of model compounds for the microstructure analysis of poly(vinyl glycoside)s

A series of glycosylated diol and triol derivatives was synthesized in order to serve as model compounds for the analysis of the stereochemistry, regiochemistry, and defect structures of poly(vinyl glycoside)s. 13C NMR spectroscopic analysis of these compounds revealed that the attached chiral carbohydrate substituents induced a strong correlation of the chemical shifts of both the anomeric C and the alpha-C atom of the aglycon with the absolute configuration of the latter. The influence of the stereoconfiguration of beta- and gamma-C atoms as well as the regiochemistry of the aglycon on the chemical shifts of the alpha-C and the anomeric C atom was also investigated. Synthesis and Absolute Configuration of Natural 2-Pyrones

2-Pyrones are frequently produced by microorganisms and often exhibit interesting bioactivities. Therefore, a short and easy synthetic access to these natural products is desirable. Synthetic routes to nectriapyrone, gibepyrone A, racemic gulypyrone A, (+)-germicidin C, (ent)-desoxygermicidin C and (ent)-prolipyrone A via a modular approach are presented, allowing the assignment of the absolute configurations of the latter three chiral compounds. The method failed for the synthesis of (ent)-phomapyrone B that was thus synthesized via a different route, resulting in an assignment of the absolute configuration of natural phomapyrone B.

Structural requirements of dictyopyrones isolated from Dictyostelium spp. in the regulation of Dictyostelium development and in anti-leukemic activity

Cellular slime molds are fascinating to the field of developmental biology, and have long been used as excellent model organisms for the study of various aspects of multicellular development. We have recently isolated alpha-pyronoids, named dictyopyrones A-D (1-4), from various species of Dictyostelium cellular slime molds, and it was shown that compound 3 may regulate Dictyostelium development. In this study, we synthesized dictyopyrones A-D (1-4) and their analogues, investigated the physiological role of the molecules in cell growth and morphogenesis in D. discoideum, and further verified their effects on human leukemia K562 cells. Nitrogen-containing compounds 22 and 37 strongly inhibited cell growth in K562 leukemia cells, indicating that these compounds may be utilized as novel lead compounds for anti-leukemic agents.

In-tube derivatization for determination of absolute configuration and enantiomeric purity of chiral compounds by NMR spectroscopy

We have developed an in-tube derivatization method using commercially available polymer-supported coupling agents to prepare derivatives of chiral compounds directly in NMR tube with high yield and purity. Because the method does not require any workup or purification, the configuration and enatiopurity can be quickly determined by NMR analysis for a small amount of chiral compounds, which is critical for today’s fast-paced medicinal chemistry efforts in drug discovery. The application of the method was demonstrated for the derivatization of chiral amines, alcohols, diols, amino alcohols, thiols, and carboxylic acids using various chiral derivatizing agents and coupling agents. This article also serves as a practical guide for in-tube derivatization and selection of suitable chiral derivatizing agents and coupling agents for various types of chiral compounds. Copyright
