Lewis pairs for ring-opening alternating copolymerization of cyclic anhydrides and epoxides

A simple and highly active catalytic process for ring-opening alternating copolymerization (ROAC) of cyclic anhydrides and epoxides still remains a key challenge. Herein, we have described an effective group of versatile and low-toxic zinc dicarbyl/amine Lewis pairs for the ROAC. The facile route showed a high catalytic activity (TOF ? 210 h-1 at 110 C) and perfectly alternating selectivity (>99%). An unexpected highly regioselective ring-opening of asymmetric epoxides (PO, ECH and SO) was also achieved by the combination of zinc alkyls (or aryls) and amines. Of note, deprotonation side reaction of alpha-H of anhydrides with organic bases was uncovered, and subsequently was inhibited by using nonpolar solvents and Lewis acid/base pairs. Thus, an array of polyesters was synthesized by the coupling of various anhydrides (PA, CHA, SA and NA) and epoxides (CHO, PO, ECH and SO) using the same Lewis pairs. Furthermore, variable temperature 1H NMR spectral and MALDI TOF MS analyses were performed to understand the possible mechanism and microstructure. The experimental results indicated that zwitterionic alkoxide and carboxylate intermediates alternately formed to enhance the ester repeat units in chain initiation and propagation. This work provides a simple and green catalytic strategy to prepare diversified polyesters from the ROAC process of cyclic anhydrides and epoxides with considerable catalytic activity and alternating selectivity.

The inclusion of an azaspiroketal Mannich base in the membrane targeting antitubercular 6-methoxy-1-n-octyl-1H-indole scaffold resulted in analogs with improved selectivity and submicromolar activity against Mycobacterium tuberculosis H37Rv. The potency enhancing properties of the spiro-fused ring motif was affirmed by SAR and validated in a mouse model of tuberculosis. As expected for membrane inserting agents, the indolyl azaspiroketal Mannich bases perturbed phospholipid vesicles, permeabilized bacterial cells, and induced the mycobacterial cell envelope stress reporter promoter piniBAC. Surprisingly, their membrane disruptive effects did not appear to be associated with bacterial membrane depolarization. This profile was not uniquely associated with azaspiroketal Mannich bases but was characteristic of indolyl Mannich bases as a class. Whereas resistant mycobacteria could not be isolated for a less potent indolyl Mannich base, the more potent azaspiroketal analog displayed low spontaneous resistance mutation frequency of 10-8/CFU. This may indicate involvement of an additional envelope-related target in its mechanism of action.

This work is a contribution to the molecular understanding of the thermodynamic properties of the chiral compounds. A comprehensive thermochemical study of the liquid enantiopure and racemate pairs of optically active alkyl lactates has been performed. Vapor pressures of DL-(¡À)-, L-(-)-methyl-, and DL-(¡À)-, L-(-)-n-butyl esters of lactic acid were measured by the transpiration method. The liquid phase standard molar enthalpies of formation of these esters were measured by using the high-precision combustion calorimetry. The standard molar enthalpies of vaporization of alkyl lactates at 298.15 K were derived from vapor pressure temperature dependencies. Thermochemical data of these compounds were collected, evaluated, and tested for internal and external consistency. The high-level G4 quantum-chemical method was used for mutual validation of the experimental and theoretical gas phase enthalpies of formation of alkyl lactates. A critical review of the available thermochemical data for the liquid and crystalline enantiopure and racemate pairs of optically active compounds has been performed. Useful general trends in energetics of sublimation, vaporization, and formation of optically active compounds have been revealed. This knowledge is required for evaluation of new and already available experimental data for the chiral compounds, and it can be helpful to assess volatility or feasibility of processes to separate enantiomers.
