Effect of raw material, pressing and glycosidase on the volatile compound composition ofwine made from goji berries

This study investigated the effect of raw material, pressing, and glycosidase on the aromatic profile of goji berry wine. The free-run and the pressed juice of dried and fresh goji berries were used for wine production, whereas glycosidase was applied to wine after fermentation. Dried goji berry fermented wine exhibited much stronger fruity, floral, caramel, and herbaceous odors due to higher levels of esters, beta-ionone and methionol. However, fresh berry fermented wine possessed stronger chemical notes due to higher levels of 4-ethylphenol. Pressing treatment reduced the fruity and caramel odors in these fermented wines, and fresh berry free-run juice fermented wine exhibited the least floral aroma. Glycosidase addition did not alter the aromatic composition of wines. The principal component analysis indicated that goji raw material played a primary role in differentiating the aromatic profiles of the wines due to the difference on the content of 20 esters, nine benzenes, eight aldehydes/ketones, three acids, two alcohols and six other volatiles. The content differences on isopentyl alcohol, styrene, benzyl alcohol, 1-octanol, (E)-5-decen-1-ol, 1-hexanol, and -cyclocitral resulted in the segregation of the wines with and without the pressing treatment, especially for fresh berry fermented win.

The vibrational circular dichroism (VCD) spectrum (800 to 1500 cm-1) is reported for 2-methylthietane (2MT).Fixed partial charge (FPC), atomic polar tensor (APT), and charge flow (CF) methods are used to compute the VCD spectra for both stable conformers (CH3 in the axial or equatorial orientation) for the complete range of fundamental vibrations, based on 3-21G and 6-31G* harmonic force fields.The experimental and computed absorption and VCD spectra of R-(+)-2MT are compared to the previously reported spectra of R-(-)-2-methyloxetane.The success of the APT and CFmodels for reproducing the dominant ring stretching IR and VCD features of that compound is rationalized by a comparison of APT and FPC intensity expressions.Further comparison of the APT and FPC calculated electric dipole moment derivatives are interpreted to suggest values for the diagonal CS and CH stretching charge flow terms for 2MT.These investigations then provide a rationale for previously reported successes of the FPC model, and permit identification of specific vibrational modes of 2MT that are amenable to FPC-VCD modeling.The previously established conformational mixture of ca. 30percent axial and 70percent equatorial is shown to be in accord with FPC-VCD predictions for the most appropriate (beta-CH2 wagging) mode. We have assembled a set of combinatorial libraries of phosphoramidates for pharmacological evaluation. A range of functionalized and unfunctionalized diols, representing a variety of diversity elements, were converted into their corresponding dimethoxytrityl H-phosphonate derivatives which were coupled to each other to produce H-phosphonate dimers and trimers. The H-phosphonate diesters were converted into phosphoramidates by reaction with a wide range of primary and secondary amines. Very large libraries (theoretically, in excess of one million compounds) possessing five sites of diversity were generated for use in our drug discovery program. Smaller libraries with lower molecular weights were also prepared in which only two monomeric units were coupled together and converted into their phosphoramidate derivatives. Methods for the attachment of both radioactive and nonradioactive labels, including 32phosphorus, tritium, and fluorescein, have been developed. Representative single sequences were also prepared and their chemical properties studied.
