Base-catalyzed C?C cross coupling of secondary alcohols and aryl-aldehydes was achieved, when an alcoholic solution of an aryl-aldehyde was stirred under reflux for 45?h in the presence of a catalytic (20?mol%) amount of K2CO3. The consistent formation of alpha,alpha?-bis-(benzylidene) alkanones was obtained in moderate to good yields using various secondary alcohols and substituted aryl-aldehydes. Herein, alpha,alpha?-bis-(benzylidene)alkanones, which are the classical products of Claisen-Schmidt (cross aldol) condensation, have been synthesized via an alternative strategy using secondary alcohols. Bis-(benzylidene) alkanones are an integral part of various drug regimes and the production of bis-(benzylidene) alkanones without using any precious metal is a major outcome of the present reaction.

Regio- and stereoselective glucosylation of diols by sucrose phosphorylase using sucrose or glucose 1-phosphate as glucosyl donor

Previously it has been shown that glycerol can be regioselectively glucosylated by sucrose phosphorylase from Leuconostoc mesenteroides to form 2-O-alpha-d-glucopyranosyl-glycerol (Goedl et al., Angew. Chem. Int. Ed. 47 (2008) 10086-10089). A series of compounds related to glycerol were investigated by us to determine the scope of the alpha-glucosylation reaction of sucrose phosphorylase. Both sucrose and glucose 1-phosphate (G1P) were applied as glucosyl donor. Mono-alcohols were not accepted as substrates but several 1,2-diols were readily glucosylated, proving that the vicinal diol unit is crucial for activity. The smallest substrate that was accepted for glucosylation appeared to be ethylene glycol, which was converted to the monoglucoside for 69%. Using high acceptor and donor concentrations (up to 2.5 M), sucrose or G1P hydrolysis (with H2O being the ‘acceptor’) can be minimised. In the study cited above, a preference for glucosylation of glycerol on the 2-position has been observed. For 1,2-propanediol however, the regiochemistry appeared to be dependent on the configuration of the substrate. The (R)-enantiomer was preferentialy glucosylated on its 1-position (ratio 2.5:1), whereas the 2-glucoside is the major product for (S)-1,2-propanediol (1:4.1). d.e. ps of 71-83% were observed with a preference for the (S)-enantiomer of the glucosides of 1,2-propanediol and 1,2-butanediol and the (R)-enantiomer of the glucoside of 3-methoxy-1,2-propanediol. This is the first example of stereoselective glucosylation of a non-natural substrate by sucrose phosphorylase. 3-Amino-1,2-propanediol, 3-chloro-1,2-propanediol, 1-thioglycerol and glyceraldehyde were not accepted as substrates. Generally, the glucoside yield is higher when sucrose is used as a donor rather than G1P, due to the fact that the released phosphate is a stronger inhibitor of the enzyme (in case of G1P) than the released fructose (in case of sucrose). Essentially the same results are obtained with sucrose phosphorylase from Bifidobacterium adolescentis.

Optimized whole cell biocatalyst from acetoin to 2,3-butanediol through coexpression of acetoin reductase with NADH regeneration systems in engineered Bacillus subtilis

BACKGROUND: 2,3-Butanediol (2,3-BD) has a wide range of applications in chiral molecular synthesis, biofuel additives, and in food flavor additive manufacturing. Fermentation is a favorable method for 2,3-BD production. However, it requires much time and produces several NADH related byproducts which compete with 2,3-BD production. Bacillus subtilis has an excellent ability for 2,3-BD production by biocatalysis. However, its production is limited by low intracellular NADH and the reversible property of acetoin reductase (AR/2,3-BDH). The whole cell biocatalyst process with two different NADH regeneration systems was designed for efficient production of 2,3-BD in B. subtilis 168. RESULTS: Formate dehydrogenase and glucose dehydrogenase for NADH regeneration were successfully co-expressed with acetoin reductase in B. subtilis 168. After optimization of biocatalyst bioconversion conditions, B. subtilis 168/pMA5-bdhA-HpaII-fdh yielded 74.5 g L?1 of 2, 3-BD with 9.3 g L?1 h?1 productivity by fed batch and 115.4 g of 2,3-BD was achieved using same batch bacterium by three repeated batch bioconversions. On the other hand, 63.7 g L?1 of 2, 3-BD was produced with 7.92 g L?1 h?1 productivity by B. subtilis 168/pMA5-bdhA-HpaII-gdh. To our knowledge, the volume productivity obtained here is the highest ever reported for biocatalysis. CONCLUSION: A higher productivity of 2,3-BD from acetoin was achieved by whole cell biocatalysis with NADH regeneration systems in B. subtilis 168. This approach can be applied for NADH related bio-based chemicals production to improve titer, yield and productivity.

Nickel-catalyzed alkylation and transfer hydrogenation of alpha,beta-unsaturated enones with methanol

Complexes of the type [{(dippe)Ni} ( 2-Calpha,Cbeta- 1,4-dien-3-one)] (dippe = 1,2-bis(diisopropylphosphino)- ethane); n= 1, 2; enone = aromatic 1,4-pentadien-3-ones) were synthesized. The “[(dippe)Ni]” moiety derived from [(dippe)Ni(-H)]2 2-coordinated to the C,C double bonds of the corresponding alpha,beta-unsaturated enone and was fully characterized using a variety of spectroscopic techniques, for instance, single-crystal X-ray diffraction, nuclear magnetic resonance (NMR), and mass spectrometry. The complexes were assessed in a catalytic transfer hydrogenation process using methanol (CH3OH) as a hydrogen donor. This alcohol turned out to be a very efficient reducing and alkylating agent of 1,4- pentadien-3-ones, under neat conditions. The current methodology allowed the selective reduction of C=C bonds in alpha,beta- unsaturated enones to yield enones and saturated ketones by a homogeneous catalytic pathway, whereas by a heterogeneous pathway, the process leads to the formation of mono- and dimethylated ketones. In the latter case, the occurrence of nickel nanoparticles in the reaction media was found to participate in the catalytic alkylation of such dienones.

This article describes the palladium-catalysed three-component coupling of 1,3-dicarbonyl compounds with nucleophiles and propargylic electrophiles for the generation of quaternary all-carbon centres in a single step, which necessitates the simultaneous control of regio-, chemo- and enantioselectivity. The use of propargyl enol carbonates, the source of two of the components, was found to be essential in maintaining high levels of regiocontrol and chemoselectivity, whereas a careful analysis of p K a trends of O-, C- and N-nucleophiles as the other coupling partner indicates that the highest levels of selectivity are likely to be obtained with relatively acidic species, such as phenols, 1,3-dicarbonyl compounds and aromatic N-heterocycles. Finally, studies towards the development of the catalytic enantioselective construction of quaternary all-carbon centres by means of alkenylation and allylic alkylation are disclosed.

Syntheses and fully diastereospecific photochromic reactions of thiophenophan-1-enes with chiral bridges

Thiophenophan-1-enes with chiral polyether bridges were prepared and their diastereospecific photochromic reactions were studied. The coupling reaction of substituted dithienylethenes and various chiral synthons afforded thiophenophan-1-enes, namely, bridged dithienylethenes, as single enantiomers without optical resolution, thus indicating that these reactions occurred diastereoselectively. Upon UV irradiation, each optically active thiophenophan-1-ene isomerized to the corresponding enantiomer of the closed form and returned to the initial enantiomer of the open form upon visible irradiation. Because thiophenophan-1-enes never isomerized to other diastereomers even at a high temperature, they underwent diastereospecific photochromic reactions. Large changes were observed in the measurement of the optical rotations of the solutions of thiophenophan-1-enes at 588 nm according to their photochromic reactions. As there was no absorption at this wavelength for both isomers of each thiophenophan-1-enes, the nondestructive readout of the photochromic reaction could be carried out by using these chiral thiophenophan-1-enes. Photo finish: Photochromic reactions of thiophenophan-1-enes with chiral polyether bridges occurred completely in a diastereospecific manner, even at high temperatures.
