Two series of new enantiopure bidentate bis(diamidophosphite) ligands with diazaphospholidine and diazaphosphepine heterocyclic backbones were prepared. The ligands have a highly modular structure, which is well suited to the synthesis of a small library of compounds. Preparation was accomplished by the successive addition of enantiomerically pure substituted diamines (N,N?-dibenzylcyclohexane-1,2-diamine (1), N,N?-dimethylcyclohexane- 1,2-diamine (2), and N,N?-dimethyl-1,1?-binaphthyl-2,2?- diamine (3)) and enantiomerically pure diols (butanediol (a), cyclohexanediol (b), di-O-isopropylidenethreitol (c), and binaphthol (d)) to phosphorus trichloride. The corresponding bis(diamidophosphite) selenides were prepared, and the 1JPSe values were calculated in order to evaluate the sigma-donor ability of the new ligands. The cationic Rh(I) complexes [Rh(COD)(P,P)]BF4 were synthesized with 8 of the 12 new bis(diamidophosphite) ligands. The complexes were used as catalytic precursors for the asymmetric hydrogenation of benchmark substrates, namely methyl alpha-acetamidoacrylate (4), methyl (Z)-alpha-acetamidocinnamate (5), and dimethyl itaconate (6). The influence of the nature of both the terminal and bridging fragments of the bis(diamidophosphite) ligands on the asymmetric induction is discussed. Most proved to be effective catalysts for the process, attaining total conversion and excellent enantioselectivity (>99% ee) with the complex containing the (R;Ral,Ral;R)-3c ligand in the hydrogenation of the three substrates. The best performing catalytic precursor [Rh(COD)((R;Ral,Ral;R)-3c)]BF4 was tested in the hydrogenation of selected cyclic enamides (7-9) and beta-acetamidoacrylate (10).

Identification and characterization of a mycobacterial NAD+-dependent alcohol dehydrogenase with superior reduction of diacetyl to (S)-acetoin

An enzyme capable of reducing acetoin in the presence of NADH was purified from Mycobacterium sp. B-009, a non-clinical bacterial strain of soil origin. The enzyme is a homotetramer and can be classified as a medium-chain alcohol dehydrogenase/ reductase based on the molecular weight of the monomer. Identification of the structural gene revealed a limited distribution of homologous genes only among actinomycetes. In addition to its activity as a reductase specific for (S)-acetoin (EC 1.1.1.76), the enzyme showed both diacetyl reductase (EC 1.1.1.304) and NAD+ -dependent alcohol dehydrogenase (EC 1.1.1.1) activities. (S)-Acetoin and diacetyl reductases belong to a group of short-chain alcohol dehydrogenase/reductases but do not have superior abilities to dehydrogenate monoalcohols. Thus, the purified enzyme can be readily distinguished from other enzymes. We used the dual functionality of the enzyme to effectively reduce diacetyl to (S)-acetoin, coupled with the oxidation of 1-butanol.

The vibrational circular dichroism (VCD) spectrum (800 to 1500 cm-1) is reported for 2-methylthietane (2MT).Fixed partial charge (FPC), atomic polar tensor (APT), and charge flow (CF) methods are used to compute the VCD spectra for both stable conformers (CH3 in the axial or equatorial orientation) for the complete range of fundamental vibrations, based on 3-21G and 6-31G* harmonic force fields.The experimental and computed absorption and VCD spectra of R-(+)-2MT are compared to the previously reported spectra of R-(-)-2-methyloxetane.The success of the APT and CFmodels for reproducing the dominant ring stretching IR and VCD features of that compound is rationalized by a comparison of APT and FPC intensity expressions.Further comparison of the APT and FPC calculated electric dipole moment derivatives are interpreted to suggest values for the diagonal CS and CH stretching charge flow terms for 2MT.These investigations then provide a rationale for previously reported successes of the FPC model, and permit identification of specific vibrational modes of 2MT that are amenable to FPC-VCD modeling.The previously established conformational mixture of ca. 30percent axial and 70percent equatorial is shown to be in accord with FPC-VCD predictions for the most appropriate (beta-CH2 wagging) mode. In-Situ Monitoring of Enantiomeric Excess during a Catalytic Kinetic Resolution

Vibrational Circular Dichroism combined with FTIR spectroscopy (VCD-IR) is demonstrated as a viable tool for the in situ measurement of enantiomeric excess during asymmetric catalytic transformations. Employing the Jacobsen (salen)Co-catalyzed hydrolytic kinetic resolution of racemic epoxides as a proof-of-concept case study, methodology is developed to monitor the enantiomeric excess of the epoxide substrate as a function of conversion of the limiting reactant, water. Comparison of results for monomeric and oligomeric catalysts probes the molecularity of the catalyst by investigating nonlinear effects in catalyst enantiopurity. These results are in excellent agreement with previous mechanistic investigations of this reaction based on kinetic measurements and computational studies.
