Stereoselective Acetalization of 1,3-Alkanediols by l-Menthone: Application to the Resolution of Racemic 1,3-Alkanediols and to the Determination of the Absolute Configuration of Enantiomeric 1,3-Alkanediols

A general and reliable method for the resolution of racemic 1,3-alkanediols, which involves their conversion into diastereomeric spiroacetals derived from l-menthone, is described.Thus, the reaction of the bis-O-trimethylsilyl derivatives of racemic 1,3-alkanediols with l-menthone in the presence of a catalytic amount of trimethylsilyl trifluoromethanesulfonate affords the diastereomeric spiroacetals 3 and 4.The two can be readily separated by silica gel column chromatography.Hydrolysis of each diastereomer under acidic conditions liberates the corresponding enantiomerically pure diol.An empirically derived correlation of configuration and 1H NMR chemical shifts for spiroacetals 3 and 4 has been developed which is rationalized based on long-range effects due to the magnetic anisotropy inherent to the menthane ring in a rigid spiroacetal conformation.The method described here should be widely applicable to the determination of the absolute configuration of various 1,3-alkanediols.

Two series of new enantiopure bidentate bis(diamidophosphite) ligands with diazaphospholidine and diazaphosphepine heterocyclic backbones were prepared. The ligands have a highly modular structure, which is well suited to the synthesis of a small library of compounds. Preparation was accomplished by the successive addition of enantiomerically pure substituted diamines (N,N?-dibenzylcyclohexane-1,2-diamine (1), N,N?-dimethylcyclohexane- 1,2-diamine (2), and N,N?-dimethyl-1,1?-binaphthyl-2,2?- diamine (3)) and enantiomerically pure diols (butanediol (a), cyclohexanediol (b), di-O-isopropylidenethreitol (c), and binaphthol (d)) to phosphorus trichloride. The corresponding bis(diamidophosphite) selenides were prepared, and the 1JPSe values were calculated in order to evaluate the sigma-donor ability of the new ligands. The cationic Rh(I) complexes [Rh(COD)(P,P)]BF4 were synthesized with 8 of the 12 new bis(diamidophosphite) ligands. The reaction of Co(eta3-C8H13)(eta4-C8H12) and Pt2(dba)3 (dba = bis-dibenzylidene acetone) under dihydrogen leads in the presence of poly(vinylpyrrolidone) to nanometric bimetallic particles CoxPt1-x. The composition of the material is determined by the initial ratio of the two organometallic precursors. The bimetallic character of the particles is evidenced by magnetic, structural, and spectroscopic studies. Structural changes are evidenced both by HREM and WAXS: platinum rich particles adopt a fcc crystalline structure while cobalt rich particles adopt a non periodic polytetraedral arrangement. The magnetic properties of the materials have been inves-tigated. ZFC/FC measurements show that the particles are superparamagnetic and that their size dispersity is very low. From isothermal magnetization measurements, the extrapolated value of the coercive field HC (T = 0) is shown to increase continuously with Pt concentration, which is characteristic of bimetallic particles.

A simple and high-yielding route to tough polyarylenes of the type poly(meta,meta,para-phenylene) (PmmpP) is developed. PmmpP is tough even in its as-synthesized state which has an intermediate molar mass of Mw?60 kg mol?1 and exhibits outstanding mechanical properties at further optimized molecular weight of Mw=96 kg mol?1, E=0.9 GPa, ?=300 %. Statistical copolymers with para,para-spiropyran (SP) are mechanochromic, and the toughness allows mechanochromism to be investigated. Strained samples instantaneously lose color upon force release. DFT calculations show this phenomenon to be caused by the PmmpP matrix that allows build-up of sufficiently large forces to be transduced to SP, and the relatively unstable corresponding merocyanine (MC) form arising from the aromatic co-monomer. MC units covalently incorporated into PmmpP show a drastically reduced half life time of 3.1 s compared to 4.5 h obtained for SP derivatives with common 6-nitro substitution.

Catalytic signal amplification for the discrimination of ATP and ADP using functionalised gold nanoparticles

Diagnostic assays that incorporate a signal amplification mechanism permit the detection of analytes with enhanced selectivity. Herein, we report a gold nanoparticle-based chemical system able to differentiate ATP from ADP by means of catalytic signal amplification. The discrimination between ATP and ADP is of relevance for the development of universal assays for the detection of enzymes which consume ATP. For example, protein kinases are a class of enzymes critical for the regulation of cellular functions, and act to modulate the activity of other proteins by transphosphorylation, transferring a phosphate group from ATP to give ADP as a byproduct. The system described here exploits the ability of cooperative catalytic head groups on gold nanoparticles to very efficiently catalyze chromogenic reactions such as the transphosphorylation of 2-hydroxypropyl-4-nitrophenyl phosphate (HPNPP). A series of chromogenic substrates have been synthesized and evaluated by means of Michaelis-Menten kinetics (compounds 2, 4-6). 2-Hydroxypropyl-(3-trifluoromethyl-4-nitro)phenyl phosphate (5) was found to display higher reactivity (kcat) and higher binding affinity (KM) when compared to HPNPP. This higher binding affinity allows phosphate 5 to compete with ATP and ADP to different extents for binding on the monolayer surface, thus enabling a catalytically amplified signal only when ATP is absent. Overall, this represents a viable new approach for monitoring the conversion of ATP into ADP with high sensitivity.
